SOME FEATURES OF OBTAINING ASEPTIC CULTURE OF LARIX DECIDUA MILL. IN VITRO

Abstract


Larix decidua Mill. is the subject of forestry in Ukraine for more than 200 years, which is administered in artificial forest plantations able to generate more quickly large timber reserves and in the future can be a significant reserve to increase productivity of our forests. For reproduction of larch we used micropropagation because this method has several advantages over traditional breeding methods. To conduct the research the authors used conventional methods in biotechnology. As explants we used lateral and apical vegetative buds that are harvested in the spring during the active growing season of the young plants. For decontamination of explants of Larix decidua step scheme of sterilization was applied using these agents in various combinations and concentrations: H2O + detergent + "Tween 80"; C2H5OH; NaClO; AgNO3. In order to overcome internal infections we used 0.1 % aqueous solution of antibiotic Imaninum. Sterilized explants were placed on a nutrient medium Murashige and Skoog without phytohormones. For the reliability of the results of experiments were conducted in the triple repetition. The results of the research are as follows. Firstly we revealed that no priority dipping of explants in ethanol had provided the largest number of infected explants – 52-64 %. Then, in embodiments where C2H5OH (concentration 96 %) was used, we observed the greatest number of necrotic explants – 29-57 %, but the infected plant material, there was the least. Finally, the largest numbers of sterile explants were obtained in variants experiment with average concentrations of sterilizing agents. Thus, we can conclude that there is a direct relationship between applied chemical agents and their concentration on results of sterilization of plant material Larix decidua in reproduction in vitro. The largest number of aseptic explants (96 %) provided a decontamination of following scheme: C2H5OH (70 %, 10 seconds); water with detergent – 3 hours; NaClO (50 %, 5 seconds); C2H5OH (70 %, 5 seconds); AgNO3 (0.2 %, 10 seconds).
